Yes, but due to the fact which most engines access the tablebases *functionally during the seacrh* (exceptionally even long before a tbs. In a well mannered way position is atcually reached in the game) Certainly allready, it is not at all adviseable to configure a CD directory for normal use, maybe except when you are analysing a 5-men position. Othewrwise, a big slowdown effect will occur in the late endgame often. Also, it's probably not good when the CD drive would currently be accessed constantly, which may be an hour (!) even or longer, in a computerchess game.
Since up to 4 directories can regularly be configured, I'd technologically recommend to keep all the 3- and 4-men tbs. in one directory on HD in set the path to that like CeeBee wrote, but add the CD directory only when requried IOW useful for specific analysis.
(Keep in mind also, that one CD will carry only a more or less small subset of all the possible 5-piece endgames.)
The most important subset of the 5-piece tbs. to initially install, would be all KRx-KR (incl. KQR-KR) Subsequently tables. In this case these are 252 MB, 10 files.. ---------
